If we see to know that some domestic animal became ill we try to know the reason for their illness and try to know disease causes.In this purpose first we go those side where wild animals are present we observe wild animals to know that symptom are present or not.
We notice following things in wild animal.
  • Not eat properly
  • Feel sleepy.
  • Slow walk.
We observe that Trypanosoma(Leopard) cause sleeping sickness.
  • First step is Sampling.

Invasive and Non Invasive 
  • Invasive Sampling.
In this method give stress to animal by use of injection syringe on vein this method is not useful.
  • Non Invasive.
With out use of syringe.Mostly use fecal matter for sampling DNA also present in fecal matter.

➽Preservation Labeling.
DNA sample preserve in liquid nitrogen to minus 80 degree centigrade.Ethanol also use for preservation.

➽DNA extraction.
In DNA extraction first mist the sample and add lysis solution incubate the sample for 8 hours next day lysate are prepared.Centrifuge the lysate 2 layer appear.

Add ethanol and again centrifuge at last add T.E buffer DNA extract from sample.
➽Gel Electrophorosis.

For confirmation of DNA do gel electro phorosis 


➽PCR
To make multiple copies of sample DNA do PCR and again AGE process repeat.

➽Sequencing

Sequencing the sample according to their complementary bases.
